High quality cDNA libraries with:

-   size fractionated DNA

-   directionally cloned in plasmid or lambda vectors

-   titers of at least 10⁶ clones

-   normalized to equalize the representation of novel and rare transcripts

-   includes sequencing of 192 clones to determine   redundancy and clone identity.

-cDNA libraries are subtracted for enrichment for low abundance transcripts and/or to eliminate clones that are commonly expressed in other tissues.

-E.coli-free DNA from BAC clones or high quality genomic DNA is randomly sheared to a size of 1.5kb to 5kb, double adaptored to avoid chimeric clones and cloned into plasmid vectors for easy -and highly successful sequencing.
